Deneke, Anneliese (University of Wisconsin, Madison) and Hans Blobel. Fibrinogen media for studies on staphylococci. J. Bacteriol. 83:533-537. 1962-Fibrinogen media were prepared simply by spreading rabbit plasma or a bovine fibrinogen solution over the surface of certain solidified agar media, thus eliminating the necessity of incorporation of fibrinogen into liquefied agar media at a critical temperature. With agar media composed of heart infusion broth, blood agar base, or Trypticase soy broth, distinct coagulase reactions were observed which corresponded closely to those obtained with the tube test.A bovine fibrinogen solution, applied to the surface of some of the commonly used selective media for staphylococci, provided an additional and more specific test system for coagulase production of individual colonies, and consequently aided in the presumptive identification of pathogenic staphylococci.

Interaction of human fibrinogen with staphylococci: presence of a binding region on normal and abnormal fibrinogen variants and fibrinogen derivatives.
Human fibrmnogen interacts with pathogenic staphylococci, causing their clumping through an unexplained mechanism. We attempted to characterize the interaction between the staphylococcal fibrinogen receptor(s) and the binding region on the fibrmnogen molecule by employing binding studies and the clumping reaction. Binding studies revealed that the Aa chain of human fibrinogen possesses a high a...

" Coagulase Plates" When human blood bank plasma is mixed with nutrient agar in a concentration of about 12 to 15O% v v and poured into Petri dishes at a temperature of 45° C. the mixture sets as a clear gel. Staphylococci inoculated on to this medium develop opaque haloes around the colonies in 12 to 48 hours if they produce coagulase (Lack and Wailling, 1954). The advantages of the plate meth...
متن کاملFibrinogen-binding protein/clumping factor from Staphylococcus aureus.
The binding of staphylococcal components to fibrinogen was studied. Fibrinogen-binding material from lysed staphylococcal cells or culture supernatants was affinity purified on fibrinogen-Sepharose and analyzed on Western (immuno-) blots by the use of fibrinogen and antifibrinogen antibodies. Two main bands of 87 and 19 kilodaltons (kDa) and a weaker band of 35 kDa bound specifically to fibrino...
متن کاملInhibition of fibrinogen reaction by polysaccharide of encapsulated Staphylococcus aureus.
Encapsulated and nonencapsulated strains of Staphylococcus aureus which lack coagulase or clumping factor (bound coagulase), or both, were examined for the antigen associated with the fibrinogen-cell clumping reaction. Extracts of the cells were tested for the ability to react with fibrinogen or to inhibit fibrinogen precipitation. Antisera prepared against encapsulated (coagulase-positive, clu...
متن کاملThe production and testing of staphylococci with clumping factor activity for use in the assay of fibrinogen degradation products.
A method for the large-scale production of a staphylococcal preparation for use in the assay of fibrinogen degradation products is described. The material is assayed against a fibrinogen standard and is shown to be stable over long periods and of high sensitivity. The possibility of production on an occasional basis in a routine laboratory is discussed.
